In order to attain Anima in Baaj temple, all the destruction spheres must be used in the cloister of trials to unlock the chests that contain the hidden items. Another reason to properly complete the cloister of trials is the useful items that will help you along the way.

Touch the Glyph, then the wall next to it to open the door to the Besaid Cloister of trials
Besaid cloister of trials
Go down, get the glyph sphere
Besaid cloister of trials
Insert glyph sphere on the door
Besaid cloister of trials
Take the glyph sphere from the door
Besaid cloister of trials
Insert glyph sphere in the first slot across the hall, inside is a Destruction Sphere, don’t take it now
Besaid cloister of trials
Touch symbols in front of the pedestral on the other side of the hall
Besaid cloister of trials
Take the Besaid Sphere inside
Besaid cloister of trials
Insert Besaid Sphere on pedestal
Besaid cloister of trials
Go back to the Destruction Sphere
Besaid cloister of trials
Insert Destruction Sphere where the Besaid Sphere was before
rod-of-wisdom
Get Rod of Wisdom on the end of the hall, the requirement to get the Aeon Anima
Push the pedestal into the marked spot on the floor to complete the Besaid Cloister of Trials.
